Use AFS-S If your subject is standing still (portraits) Use AFS - C if your subject is moving ( Sports

Not sure if you are asking What is Metering or which metering mode.If metering Mode, Use Matrix unless you are more advanced and know why not to

Press the shutter Button half way to start metering ( You must be on AFS-S to do this) with the shutter button pressed half way this will lock the focus on your subject. Keeping you finger press half way (DON'T let up) recompose your framing and press the shutter completely to fire

Save JPEGs as Large and Fine Quality
 
You really, really, really need to read through the camera manual. All that you asked is covered there.

Yeah, not to be a smart alec, but you should read these things up (camera in hand).

The focusing thing is simple, focus on your subject, hold the shutter button down half-way (which will lock in focus and exposure) and then recompose the shot.
