Photo tagging - developing a workflow

As a location scout, I have a 35K + database, growing rapidly. I use ACDSee on my desktop and SmugMug as an online database. Photo tagging is getting more and more critical. Search has to be made within minutes, online and offline.
ACDSee has a very powerful database, but it uses a proprietary format and I am worrying about this. It is very easy to use while on my desktop, and very powerful, but any proprietary format makes me nervous and I am thinking about switching for a more universal format that SmugMug and other softwares can read.

I am sure many of you have large databases. What do you use for tagging? Have you find a system that works both in offline programs and online galleries?
