Either one. If you're shooting a static subject, ( landscape/cityscape, architecture) Manual works fine. If you're shooting a fast-moving subject, (sports, kids playing), go to Aperture preferred.

I'm thinking you are talking about the 'm' and 'a' on the lens body itself. Correct me if I'm wrong.

That switches the lens from 'autofocus' to 'manual focus'. It has no direct effect on the aperture you selected for the image.
